serial sectioning in the sem for three dimensional

In the present work, we report our recent progress in the development, optimization, and application of a technique for the three-dimensional (3-D) high-resolution characterization of crystalline microstructures. The technique is based on automated serial sectioning using a focused ion beam (FIB) and characterization of the sections by orientation microscopy based on electron backscatter

After SEM observation, the samples were softened with 2% NaHCO3 and observed with histological serial sectioning. By combining both techniques, that is combined analysis of outer structures for SEM and inner structures for serial sectioning, three dimensional analysis of the colonic adenoma and carcinoma in adenoma were successfully completed.

A method of accurate serial sectioning has been designed for three-dimensional texture measurement in aluminium alloy sheet. Using this method, determination of three dimensional texture in an AA6111-T4P sheet has been carried out by electron backscatter diffraction (EBSD) in a scanning electron microscope (SEM) from the sheet surface to 1/3 of the thickness.

Serial Section Scanning Electron Microscopy (S3EM) on

2012/4/16High resolution, three-dimensional (3D) representations of cellular ultrastructure are essential for structure function studies in all areas of cell biology. While limited subcellular volumes have been routinely examined using serial section transmission electron microscopy (ssTEM), complete ultrastructural reconstructions of large volumes, entire cells or even tissue are difficult to achieve

Another serial-sectioning SEM method uses a focused ion beam (FIB) instead of a microtome in a specimen chamber (FIB-SEM; Knott et al., 2008). The FIB system etches (mills) away atoms on the specimen surface with a gallium ion (Ga + ) beam, and is a commonly used micro- and nano-machining tool in the semi-conductor industry and materials science.

In this study, a serial sectioning technique was employed in order to visualize the three-dimensional (3D) structure, and to accurately simulate the mechanical and thermal behaviors of SiC particle-reinforced Al composites. Sequential, two-dimensional (2D) optical images of the microstructure were acquired after polishing, and then reconstructed to develop 3D geometries for microstructural

In this work, a three-dimensional (3D) microscopic elemental analysis system based on a serial sectioning technique was developed to observe the internal structure of such materials. This 3D elemental mapping system included an X-ray fluorescence analyzer and a high-precision milling machine.

We reported our observation results on the etch pit shapes on β-Ga2O3 (001) wafers using a scanning electron microscope (SEM) and an atomic force microscope (AFM) in a previous study. However, it was difficult to detect the internal crystal defects that exist under the etch pits. In this study, to gain a detailed understanding of the internal crystal defects under the etch pits in detail, we

Serial section scanning electron microscopy

In contrast to FIB/SEM and SBF/SEM, serial-section SEM allows the sections to be immobilized on the glass slide. For this reason, the sections can be stained with heavy metals. Here, the sections are stained with a 1% aqueous solution of uranyl acetate for 10min, rinsed with distilled water, and then stained with Reynolds lead solution for 5min (Fig. 2D).

For serial block-face SEM, the embedded samples are trimmed, mounted on the specimen pins and coated with gold metal to increase electron conductivity prior to the SEM imaging (d and e). All the procedure from fixation to embedding usually takes about 2–3 days, while trimming and mounting of the block specimens take less than 1day.

In this chapter we describe three different approaches for three-dimensional imaging of electron microscopic samples: serial sectioning transmission electron microscopy (TEM), scanning transmission electron microscopy (STEM) tomography, and focused ion beam/scanning electron microscopy (FIB/SEM) tom

Three‐dimensional SEM

For the past 50 or so years, the ability to obtain extensive three‐dimensional information from cells has been the preserve of those brave electron microscopists willing to undertake extensive serial thin sectioning followed by manual or semiautomated reconstruction.

For this chapter we describe serial sectioning TEM, STEM tomography, and FIB/SEM tomography. The most important prerequisite for three-dimensional imaging is good specimen preparation. For all these three imaging methods, cryo-fixed (high-pressure frozen) and freeze-substituted samples are used.
